Introduction. Several laticifer plants are known to have significant amount of proteases in their latex. These proteases can be used in the food industry in order to reduce the allergenicity, increase the digestibility, or obtain hydrolysates to specific diets. Furthermore, these hydrolysates can generate different bioactive peptides released during hydrolytic reactions.ObjectivesThe aim of this study was to evaluate the biological activity of the bovine casein hydrolysates by latex proteases of five different species.Material and MethodsThe proteolytic fractions of the latex of Calotropis procera (CP), Cryptostegia grandiflora (CG), Plumeriarubra (PR), Himatanthusdrasticus (HD) andThevetiaperuviana (TP) were extracted and the caseinolytic activity was measured. The hydrolysis of the bovine casein was performed for 30 and 120 min at 45 °C and the hydrolysis degree was determined by TNBS method. The casein hydrolysatewas analyzed by Tricine-SDS-PAGE and the peptide concentration was determined by Lowry?s method. The antioxidant activity was determined according toâ-carotene and DPPH assays, and the Angiotensin I converting enzyme (ACE) inhibitory activity was also measured.Results and DiscussionAmong all the studied species, CP, CG and TP showed higher caseinolytic activity: 1.03 ± 0.03; 0.89 ± 0.01 and 0.85 ± 0.04 AU/ml, respectively. The latex of PR and HD did not show caseinolytic activity. The hydrolysis degree (%) of the casein by latex proteases was 14.3 ± 0.5 (CP), 17.3 ± 2.3 (CG), and 21.2 ± 2.5 (TP). The antioxidant activity was slightly positive in the hydrolysate obtained using CP (120 min). ACE inhibitory activity was only detected in the hydrolysates obtained using CG (120 min) (>10% inhibition).ConclusionsThe latex of CP and CG are important resource of proteases that can be used not only to hydrolysate the casein but also in the industry as ingredients in the food processing.
