The Arenaviridae family virions are enveloped and contain tow segmented of ambisense single stranded RNA. The purpose of this work is to analyze the main internalization mechanism of Tacaribe virus (TCRV) in cells that differentially express the human transferrin receptor (hTfR 1). We treated cells with different concentrations of chlorpromazine (drag that inhibit the clathrin-dependent endocytosis) and nystatine (drag that inhibit the cholesterol-dependent endocytosis) and analyze the number of infected cells by indirect immunofluorescence. We also transfect this cells with dominant negative variant of cellular proteins involved in clathrin-dependent endocytosis and analyze the percentage of infected cells by direct immunofluorescence. The results show that in cells with no expression of hTfR 1, TCRV use the clathrin-dependent endocytosis but in cells that express the hTfR 1, the main entry pathway is the cholesterol-dependent pathway.
