Traditional methods for drug-susceptibility testing (DST) are slow, laborious and costly, which indicates the need for a new diagnostic method that combines sensitivity, specificity, and speed. Our goal is to evaluate the performance of fluoromycobacteriophages directly in sputum samples. Methods: We tested the performance of mCherrybombΦ in tuberculosis negative sputum samples spiked with M. tuberculosis mc26230, samples were processed. After infection, the cells were fixed with 4% PFA  and can be detected using a fluorescence microscope. Different parameters were evaluated in order to determine the optimal conditions to obtain satisfactory results (pH, liquefaction and decontamination protocols, timing of infection). Clinical samples were tested and evaluated the ability to discriminate between strains resistant to rifampicin Results: We have developed a second generation of fluoromycobacteriophages with higher sensitivity and a shorter time to detection of signal. These new reporter phages can detect M. tuberculosis and discriminate between rifampicin resistant and sensitive strains directly in sputum samples, getting results in just 24-48 hours from the sampling. Conclusions: Close to our goal, a two day assay directly in sputum samples was developed. Our current research will result in the development of a simple, rapid and inexpensive method that will determine the presence and pattern of drug resistance of Mycobacterium tuberculosis directly from sputum samples in a short time, providing therapy and preventing the spread of drug-resistant strain.
