MINIMUN IHIBITORY CONCENTRATION (MIC) OF ANTI- Y. enterocolitica BIOFILMS ALKALOIDS.

Resumen:
The biofilm mode of growth results in an increased bacterial resistance against classical antimicrobial treatment. Thus, the search for alternative therapies is a necessity. Previously a screening was performed among 16 different alkaloids and four compounds were selected for their ability to inhibit Yersinia enterocolitica (Ye) biofilm formation at a concentration of 500 microM; so the aim of this study was to determinate the minimum inhibitory concentration (MIC) of these alkaloids against Ye B1A/O:7,8-8-1,9 (CLO001) strain. This strain was selected for its high ability to form biofilms onto polystyrene. The culture medium was trypticase soy broth (TSB) added with 0.25% glucose and the biofilm formation was determined by the crystal violet technique after 24 h of incubation at 25°C onto polystyrene-96 well plates. The initial concentration of each compound was 500 microM and serial dilution 1/2 were performed. Sessile cells were measure at 550 nm. With compounds 3 and 10 the MIC observed for was 250 microM and with compounds 11 the MIC was 31.25 microM. On the other hand, the most effective compound was number 6 with a MIC of 3.9 microM. Therefor, these kinds of compounds (primary number 6) could be use as new strategies to control microorganisms, specially the resistant biofilms.
