Resumen:
T cell exhaustion is one of the mechanisms that accounts for immunosuppression on the tumor microenvironment. It is characterized by a sustained overexpression of inhibitory receptors and a progressive loss of effector functions. The aim of this study was to investigate the biogenesis and exocytosis of cytotoxic granules during CD8 T cell exhaustion, focusing on the role of O-GlcNAcylation as regulator of these processes. To address this, we purified splenic CD8 T cells from C57/BL6 mice and exposed them to an activation/reactivation cycle using anti CD3/CD28 to generate exhausted CD8 T cells (CD8ex).
