Cyclic Modulation of the Actin Filament Network Associated to Sertoli- Sertoli Junctional Complexes in the Seminiferous Epithelium of the Rat. J. Cavicchia1; F. Capani1; M. Fóscolo1; A. Morales1; A. Histology and Embryology, Cuyo Medical School, Mendoza, Argentina.   Using several immunomicroscopic techniques combining beta actin with prosaposine and glutaredoxine antibodies (to mark Sertoli organelles) we observed the structural ( confocal microscopy) or ultra structural ( electron microscopy) organization of tight and adherents junctions, which are the morphologic substrate of the blood testis barrier (BTB) and also characteristic of ectoplasmic specializations found usually higher in the seminiferous epithelium. This confocal colocalization also permits to observe the intricate arrangement of the actin contained Sertoli branches, which surround germ cells in different stages of the spermatogenic cycle. Such techniques were also combined with immunoblotting of beta actin to measure this cytoskeleton protein at distinct stages of the spermatogenic cycle. It was shown a clear modulation of the actin pool and also of the actin spatial organization, before, during and after the liberation of spermatozoa in the tubular lumen (spermiation). In conclusion, significant interrelations between the actin network and the junctional complexes of the BTB were detected at different stages of the seminiferous cycle.
