EFECT OF GLP-1 IN HUMAN MESNCHYMAL STEM CELLS ADIPOGENESIS

AbstractMesenchymal stem cells are multipotent adult cells with great potential for clinical applications for their abilityto differentiate in vitro and in vivo into several cell types including osteocytes, chondrocytes, adipocytes, etc. Inthis work human adipose-derived mesenchymal stem cells (hASC) were isolated from adipose tissue voluntarilydonated by healthy plastic surgery patients who signed an informed consent approved by the National University ofCuyo ethical board. We performed a multicolor characterization by flow cytometry, which showed cell populationsin average ¿ 95% cells CD90+, CD105+ and CD73+, and ¿98% CD34-, CD11b-, CD19-, CD45- and HLA-DR-.In vitro, hASC adipogenic differentiation occurs by the addition of an adipogenic differentiation standard medium(ADSM). This includes high glucose DMEM (Gibco), 10% FBS, synthetic drugs (250 nM dexamethasone, 0.5 mMIBMX, 2 mM rosiglitazone) and peptidic hormones (10 μg/ml insulin). Our aim was to evaluate the effect of Ex-enatide, a biosimilar drug to GLP-1 (glucagon-like peptide 1) in adipogenesis of hASC. Cells were induced to theadipogenic linage using the ADSM, and the same formula but independently substituting IBMX and rosiglitazoneby 1 μg/ml GLP-1, or GLP-1 alone in 10% FBS DMEM. Untreated (control) hASCs were cultured in 5% FBS sup-plemented DMEM. The hASCs were fixed and stained with Red Oil, which is specific to dye triglycerides containedin lipid droplets. The lipid droplets content was quantified by an algorithm developed in our laboratory. Amongthe different tested drug cocktails, the replacement of IBMX by GLP-1 was the only one to be more efficient inadipogenic induction than our routinely used ADSM. The possible role of GLP-1 in this differentiation process isdiscussed.
