p { margin-bottom: 0.25cm; line-height: 120%; }Mesenchymalstem cells are multipotent adult cells with great potential forclinical applications for their ability to differentiate in vitro andin vivo into several cell types including osteocytes, chondrocytes,and adipocytes. In this work human adipose-derived mesenchymal stemcells (hASC) were isolated from adipose tissue voluntarily donated byhealthy plastic surgery patients who signed an informed consentapproved by the ethical board of the Universidad Nacional de Cuyo. Weperformed a molecular characterization by flow cytometry, whichshowed cell populations in average > 95% cells CD90+, CD105+ andCD73+, and >98 % CD34-, CD11b-, CD19-, CD45- and HLA-DR-. Invitro, hASC adipogenic differentiation occurs by the addition of anadipogenic differentiation standard medium (ADSM). This includes highglucose DMEM (Gibco), 10% FBS, synthetic drugs (250 nM dexamethasone,0.5 mM IBMX, 2 mM rosiglitazone) and peptidic hormones (10 μg/mlinsulin). Our aim was to evaluate the effect of Exenatide, abiosimilar drug to GLP-1 (glucagon-like peptide 1) in adipogenesis ofhASC. Cells were induced to the adipogenic linage using the ADSM, andthe same formula but independently substituting IBMX androsiglitazone by 1 μg/ml GLP-1, or GLP-1 alone in 10% FBS DMEM.Untreated (control) hASCs were cultured in 5% FBS supplemented DMEM.The hASCs were fixed and stained with Red Oil, which is a specificdye of intracellular triglycerides. The lipid droplets content wasquantified by an algorithm developed in our laboratory. Among thedifferent tested drug cocktails, the replacement of IBMX by GLP-1 wasthe only one to be more efficient in adipogenic induction than ourroutinely used ADSM. The possible role of GLP-1 in thisdifferentiation process is discussed.
