Effects of Dehydroleucodine on the migration and morphology of HeLa cells. Amaya C, Costantino V, Bertoldi V, Losinno A, Lopez L. IHEM-CONICET, FCM, UNCuyo, Mendoza. celinaamaya@yahoo.com.ar For the control of tumor metastasis it is important to identify chemical compounds with antimigratory potency. The present study was designed to examine the effects of a sesquiterpene lactone, Dehydroleucodine (DhL), on the migration of tumor cells in culture. For wound healing assay culture cells were grown to confluent monolayer in 12-well petri dishes. After 24 hours of incubation in serum-free medium, cells were scratched with a tip and the culture medium was replaced with medium supplemented with 1,5% FBS and hydroxyurea 2 mM (to inhibit proliferation) following the treatment of DhL at different concentration (0, 5, 10 and 20 ìM). Wounds were imaged at 0 and 24 h at the same spots by direct microscopic visualization. The cell migration percentages were for Dhl 5 ìM 94% ±1,3; 10 ìM 77% ±2,9 and 20 ìM 56% ±4,7. Wound closure by the control was set to 100%. DhL had been shown to have anti-cancer activity against HeLa cells by inhibiting cell-cycle progression. In this study, we found that DhL could effectively inhibit migration of tumor cells in vitro. DhL could be a new promising candidate as an antimetastatic agent.ìM).
