GREASE DIET PROMOTE AN INCREMENT IN SERUM AND SPERM CELL CHOLESTEROL IN ADULT MALE RABBIT GENERATING SEVERAL DAMAGES BUT VIRGIN OLIVE OIL INCORPORATION TO DIET REVERT THE NEGATIVE EFFECTS.

Specific diet prepared with 20% (w/w) of grease commercially obtained from bovine tissues promotes an increment in cholesterol serum and cells in male adult rabbit (grease groups= GG) as it was already published. But these animals did not become obese when it was compared with the control groups during the experimental time, 6 months (CG). (Body weight for GG = 4.3 ± 0.15 Kg and CG = 4.2 ± 0.2 Kg; SD > 0.5). When the fat group was divided into two batches, one was maintained with the fat diet consumed and the other with half of fat; it was found a decrease in serum cholesterol. If the diet with half fat was supplemented with olive oil , the serum cholesterol decreases to similar level of CG. Sperm cells under this environment showed several abnormalities. To further characterized the morphological changes described we obtained sperm from semen and processed for electron microscopy analyses. To investigate the hepatic tissue – the organ that regulates serum cholesterol under grease harmful or olive oil protection it was also processed liver for ultra-structural studies by conventional techniques. Longitudinal sperm head folding, cytoplasmic droplet and intracellular vacuoles were observed in abnormal sperm accompanied to GG. Hepatocytes showed big vesicles caged of lipids without a loss of organelles with normal nucleus. These cellular defects decrease considerably with virgin olive oil incorporation to diet. The molecular or cell mechanisms involved in this research are under study and need more specific effort in this way.
