The present work was designed to examine the effect of a phenolic compound purified from olive oil, hydroxytyrosol (Ht), on calcium ionophore A23187-induced rat peritoneal mast cell degranulation, with the goal of testing the hypothesis that such molecule acts as a mast cell stabilizer. Rat peritoneal mast cells were purified in Percoll and incubated with: 1) Buffer (control) or 2) A23187 or 3) Ht + A23187. â-hexosaminidase release studies by colorimetric reaction, evaluation of mast cell morphology by light microscopy (toluidine blue staining), mast cell viability (Trypan blue dye exclusion) and dose-response curves were carried out. Statistics: ANOVA/Tukey-Kramer. A23187 increased â-hexosaminidase release from rat peritoneal mast cells and elicited evident morphological changes. These effects were inhibited by 200 ìM  Ht. The present study demonstrates that Ht inhibits A23187-induced mast cell activation and increases mast cell viability, acting thus as mast cell stabilizer in rat peritoneal mast cells.
