The Effect of Dehydroleucodine in Adipocyte Differentiation

Resumen:
Dehydroleucodine (DhL) is a sesquiterpene lactone of the guaianolide groupwith gastric cytoprotective activity. 2323 Recent studies have also demonstrated that DhL inhibits the proliferation of vascular smoothmuscle cells. In this 2424 study we examined the effect of DhL in the differentiation of 3T3-L1 preadipocytes. The addition of DhL signifi- 25fi- 25 cantly inhibited the differentiation of 3T3-L1 preadipocytes alongwith a significant decrease in the accumulation 26ficant decrease in the accumulation 26 of lipid content by a dramatic downregulation of the expression of adipogenic-specific transcriptional factors 27fic transcriptional factors 27 PPARã and C-EBPá. However, phosphorylation of AMPKá, Erk1/2 and Akt1was not inhibited by DhL treatment. 28ã and C-EBPá. However, phosphorylation of AMPKá, Erk1/2 and Akt1was not inhibited by DhL treatment. 28 Interestingly, we also found that 11,13-dihydrodehydroleucodine, a derivative of DhL with inactivated á- 29á- 29 methylene-ã-lactone function, also inhibited the differentiation of 3T3-L1 preadipocytes. Taken together, these 30ã-lactone function, also inhibited the differentiation of 3T3-L1 preadipocytes. Taken together, these 30 data suggest that DhL has an important inhibitory effect in cellular pathways regulating adipocyte differentiation 3131 by modulating the PPARã expression, which is known to play a pivotal role during adipogenesisã expression, which is known to play a pivotal role during adipogenesis
