Nanocompositesarecompositesinwhichatleastoneofthephasesshowsnanometricdimensions.InthisworkweevaluatedinvivobiocompatibilityofSilicananoparticle-collagenIhydrogelscompositesasgentamicinreleasingsystemsformedicalapplications,thatshowedprolongedantibacterialactivityinapreviouswork.Onesteploaded500nmsilicananoparticles(SiNPs)weresynthesizedaccordingtotheStöbermethod.ForthispurposeTetraethylorthosilicate(TEOS)andammoniumhydroxidewereaddedtoawater-ethanolmixturewithorwithout(blank)gentamicinsulphate.MonodispersedrugfreeandgentamicinloadedSiNPswereobservedbyscanningelectronmicroscopy(SEM).CollagentypeIwaspurifiedfromrattailsandnanocompositeswerepreparedbymixing30mgSiNPswith5mg/mLcollagensuspension(0,5MSi).AntibacterialactivityofgentamicinSiNPs(338IUpergNPs)wereevaluatedbythediskdiffusionmethodasdescribedintheUSPharmacopeia.PseudomonasaeruginosawasusedassensitivemicroorganismandLBagarasgrowthmedium.CollagenhydrogelandnanocompositesmorphologywerealsoobservedbySEM.Biocompatibilitywasevaluatedinvivobyimplantingsubcutaneously300uLhydrogelorcompositeonWistarrats(250g).Nocleartoxicitywasobservedbyhistologyandimmunohistologicalstudiesafter15days.
