Resumen:
The goal of this work is to evaluate the effect of different reaction parameters on the adsorption of Doxorubicin on magnetite nanoparticles modified with folic acid (MNPs@FA). Three nominal Doxo/MNPs@FA mass ratios, the influence of pH and the incubation time were evaluated to determine the set of parameters ensuring the best formulation regarding not only the drug loading capability, but also its physicochemical properties. All the obtained samples were exhaustively characterized by FTIR, UV-V and atomic absorption (AA) spectroscopy, dynamic light scattering, z potential and TEM. A novel continous flow system based system was designed to estimate their retention ability in an environment simulating the bloodstream under the influence of an external magnet. This represents a reliable approximation to infer the amount of nanotheranostic able to reach the target site. Besides, in vivo assays were developed on Zebrafish larvae (Danio rerio) model to assess the toxicity of nanotheranostics.
