Differences in pro- and anti-inflammatory factor in kidney in a murinemodel of Hemolytic Uremic Syndrome GonzaloE. Palermo,María Victoria Ramos.HemolyticUremic Syndrome is a disease triggered by Shiga toxin (Stx) characterized byhemolytic anemia, thrombocytopenia and renal failure. Although the role ofinflammatory factors in the disease is well documented, the role ofanti-inflammatory cytokines, such as IL-10, is not clear. Previouslywe demonstrated that IL-10 deficient mice(IL-10-/-) had a higher survival after Stx2, associated with lowerrenal damage reflected by reduced plasma creatinine? levels. In this work, westudied the profile of pro- and anti-inflammatory factors in blood and kidney. Before, or3h and 24h after administration of 1LD100 of Stx2 e.v.,IL-10-/- and Wt mice were sacrificed and plasma was collected. Thekidney was isolated and the tissue was homogenized in HEPES buffer. Totalprotein concentration was measured (Bradford).The plasma (pg/ml) and renal levels (pg cytokine/mg totalprotein) of IL-10, Corticosterone, IL-6 and TNF-α  were quantified by ELISA. AlthoughIL-10 was not detected in Wt plasma, the renal level of this cytokine wasreduced at 3h in kidney of Wt mice (Wt0h:57.31±8.16/WT3h:26.72±4.47*,*<p0.05vs 0h, n=4 per group). The circulating corticosterone levels were onlyincreased in IL-10-/- mice 3h after Stx2 (IL-10-/-0h:24100±10000/IL-10-/-3h:337900±56000*#,*<p0.05 vs 0h, n=5 per group). La frase enverde me resulta rara, porque vos resaltas el descenso..pero tal vez es importanteque basalmente tienen alto.. (y el KO no tienen nada?)..o sea lo diriadiferente..ej: Although IL-10 was notdetected in Wt plasma, before or after Stx2, the renal level of this cytokinewas higher before than after Stx2 inoculation?.y/o comparar con elcomportamiento de las otras citokinas en el riñon de los wt?.que no decisnada.. (ver comentario más abajo)..por ej: es la unica detectable en el riñon???In contrast, IL-6 and TNF-α were higher at 3h after Stx, in plasma andkidney of IL-10-/- mice (IL-6 plasma: IL-10-/-0h:2.5±3.5/IL-10-/-3h:13.1±3.4;IL-6 kidney: IL-10-/-0h:0.18±0.02/IL-10-/-3h:0.30±0.11,n=2 per group; TNF-α plasma: IL-10-/-0h:2.5±2.9/IL-10-/-3h65.7±13.1*,*<p0.05vs 0h, n=5 per group; TNF-αkidney: IL-10-/-0h:72,5±2,9/IL-10-/-3h;149.0±18.44,n=2 per group).
