We have previously reported that neutrophils acquired differentprofiles of activation when they are cultured in presence of γδ Tcells activated by phosphoantigens or through CD3. Now, we aimto investigate the mechanism involved in the modulation of neutrophilsactivity by γδ T lymphocytes. γδ T cells were purified from humanperipheral blood mononuclear cells by using an anti-TCR γδMicroBead isolation kit; and neutrophils were isolated by dextransedimentation. After purification, γδ T cells were stimulated with anti-CD3 antibodies (250 ng/ml) or phosphoantigens (HMBPP, 10 μM)and cultured with or without neutrophils. After that, we analyzed inγδ T cells, IFN-γ and TNF-α production by ELISA, and CD69 expressionby flow cytometry. Also, we analyzed the activation state ofneutrophils and the expression of perforin by flow cytometry.
