Hemolytic Uremic Syndrome (HUS) is a life-threatening complication of Shiga-toxin (Stx) Escherichia coli (STEC) infections. In Argentina HUS constitutes an endemic disease. Our hypothesis is that the interaction between the mucosal associated lymphoid tissue (MALT) and STEC strains during the acute stage of infection would determine the outcome of the disease and the onset of HUS. Therefore, the aim of this work was to assess the cytokine profile induced locally in a model of HUS by intragastric administration of STEC strains in mice at weaning. In order to understand the role of Stx in the local activation of lymphocytes, we also administered the same STEC strain knocked out on stx (ΔStx) in a group of mice. To do this we removed mesenteric lymph nodes (MLN) at several times after infection (10, 12, 36, 48 and 72 h). The single cell suspensions were incubated with anti-CD3/ CD28 antibodies for 72 h. We analyzed proliferation by MTT assay and cytokine release in culture medium by ELISA (IFNγ, TGFβ, IL10, and IL17). We observed that there was a significant increased number of pre-activated lymphocytes in mice inoculated with ΔStx and STEC strains compared to control (PBS-inoculated) at 10 h after inoculation (mean OD570 ±SEM,n): Ctrol= 0.28±0.04, 6; ΔStx=0.53±0.06, 7; STEC=0.48±0.04, 14; ANOVA p
