Resumen:
The Mas receptor (MasR) is a class A Orphan G-protein-coupled receptor (GPCR). Although angiotensin-(1?7) [Ang-(1?7)] has been reported as its putative ligand, the intracellular signaling pathways activated by the MasR remain partially characterized preventing definitive knowledge of ligand/receptor pharmacology. In this study we examined MasR-dependent activation of canonical (G protein-mediated) and non-canonical (MAP kinase mediated) GPCRs? signaling pathways. With that aim, we used endogenously expressing human lung carcinoma A549 cells to evaluate Gq-coupled activity, as it was previously described by other authors, and we monitored changes in intracellular Ca2+ levels after Ang(1?7)-stimulation. Incubation with 100nM Ang(1?7) failed to increase Ca2+ levels either in endogenously expressing or MasR transfected A549 cells. On the other hand, transfection of HEK293T cells with MasR resulted in a significant decrease in basal cAMP levels (p
