Neural stem and pro​genitor cells (NSC/NPC) are multi​po​tentand have gained well-earned fame in the past decade, since theirpre​sence in the adult mammalian brain has made them one ofmany possible targets for CNS dis​ease treat​ment. We are in​ter​-ested in harnessing NSC fate spe​ci​fication in vitro as a strategyfor neuro​degenerative dis​ease cell therapies and cellreplacement therapies. We are currently studying ways toimpro​ve oligo​dendro​genesis in vitro from neo​natal micesubventricular zone primary cultures. The use of PlateletDerived Growth Factor (PDGF), together with EpidermalGrowth Factor (EGF) and basic Fibroblast Growth Factor (bFGF)is well known to favor the fate choice of multi​po​tent neuralcells towards the oligo​dendro​glial lineage. We com​pareddifferent growth factor and growth factor isoform com​binationsin vitro to generate cultures with a suitable bio​mass and highlyenriched in oligo​dendro​cyte pro​genitor cells (OPC). The PDGFhomodimeric isoforms AA and BB showed differential effects atequal con​cen​trations on OPC pro​portions after treat​ment. Thecom​bination of both these isoforms together with EGF, bFGF orEGF/bFGF was sub​stanti​ally different. Although bio​mass wascompro​mised, the best culture condition for OPC gene​ration inthis culture system was obtained by com​bining bFGF and PDGF-BB. These results are in​tended to impro​ve NSC/NPC culturetechniques when OPC-enriched primary cultures are desired.
