Background: Reactive arthritis (ReA) may develop after gastrointestinal or genitourinary infections. Dendritic cells (DCs) are central for T lymphocyte activation. We have reported that TNFRp55-/- mice develop chronic Yersinia enterocolitica (Ye)-induced ReA. On the arthritis onset (day 14), these mice presented an increase of migratory DCs (mDC:MHCIIhiCD11c+) and resident DCs (rDC:MCHIIintCD11c+) in mesenteric lymph node (MLNs) and regional lymph nodes to joints (RLN). The role of DCs in ReA is unknown. The purpose was to study the contribution of DCs migration to ReA.Methods: C57BL/6 wild-type (WT) and TNFRp55-/- mice were orally infected with 1-5× 108 Ye O:3. To block DC migration, fingolimod (FTY72) (5mg/kg) was administered intraperitoneally at days 14, 17 and 20 after infection. Clinical score was calculated until day 21, and DCs from RLNs were analyzed by flow cytometry. Bone marrow-derived DCs (BMDCs) of WT or TNFRp55-/- mice were in vitro infected with Ye O:3, and the CCR7 expression was analyzed by flow cytometry. The IL12/23p40 and IFN-γ levels were measured by enzyme-linked immunosorbent assay (ELISA) in homogenates of the joints.Results: At day 21, FTY720-treated TNFRp55-/- mice decreased their arthritis clinical score and the absolute number of mDC and rDC (CD11b+ or CD11- subsets) in their RLNs (p
