Sphingosine-1-phosphate (S1P) is a bioactive sphingolipidlined to chronic inflammation and cancer. nce produced intracellularlyby to sphingosine inases S and S, S might actas a a second messenger or b as autocrine/paracrine metaboliteby acting through proteincoupled receptors Cs, namedS. ost of S actions are mediated by its receptors,including migration, invasion and angiogenesis. reviously, ehave shon that extracellular S induces FB activation inmelanoma cell lines that do not express FilaminA FLa, anactin binding protein. Here e explored some biological actionstriggered by extracellular S in to melanoma cell lines cells FLa and A cells FLa. First, e analyzed mAexpression of Fregulated cytoines and chemoines byqC. ur results demonstrate that S stimulates mA expressionof L, TFα, CCL and CCL only in cells. yusing oydens chamber migration assays, e found that Sinduces migration in cells but the effect is minor in A cells.oreover, S protected from serum depletioninduced apoptosisin cells. n concordance ith these results, TT experimentsindicate that S preserve cell viability in cells hile its effectis less noticeable on A7 cells. In addition, gelatin degradationassays shoed that S induces secretion in cells.nterestingly, a differential behavior is observed in A cells Sinduced MMP2, but not MMP9, secretion, suggesting that FLNamay play a role in the regulation of s release. n summary, ourexperiments indicate that FLa expression modulates extracellularS inducedcytoine expression, secretion, migration andproliferation in melanoma cells.
