Abstract: CD8+ T cells contribute to the optimal control ofTuberculosis infection by inducing apoptosis on infectedmacrophages. Mtb infection is associated to alternative activationof macrophages. We aimed to study the effect of macrophageactivation on Mtb-specific CD8+ T cell induced apoptosis.Monocyte Derived Macrophages (MDMs) from healthy blooddonors were classically activated by IFN-γ or alternativelyactivated by IL-4 addition during 48 h and loaded with M.tuberculosis (Mtb) antigens. Macrophages phenotype wasdetermined by HLA-DR, CD86, DC-SIGN and CD14 staining andflow cytometry analysis. Then, MDMs were co-cultured withpreviously Mtb-stimulated, autologous CD8+ cells at differentCD8:MDM ratios. Specific killing was assessed after 3h byAnnexin V and 7-AAD staining and IFN-γ production by CD8+cells was assessed after 18h by ELISPOT assay.CD8-induced apoptosis of unactivated MDMs was Mtb-specific(p
