For the application of immobilized enzymes in nonconventional media several strategies have beenexplored. As it was described in the current chapter, from the careful selection of the supports, the definitionof the additives to be used at the immobilization step, and the posttreatment of the immobilizedenzymes, to the design of the reaction media and the use of computational tools to predict and avoidlater undesirable effects; the interactions of all the system components during reaction in nonconventionalmedia have to be adequately studied. Among different options of enzyme immobilization,CLEAs may be key for biocatalysts use in nonconventional media, but improved procedures and resultingimproved characteristics are still needed.Literature review evidences that nonconventional solvents have been very successful to give suitablereaction media for biocatalysis in several processes, especially in those involving substrates withlow solubility in aqueous solvents. By the way, there are several solvents such as DES or ILs that arewaiting for new developments to go to improved pathways of application. There is probably a bestnonconventional solvent to improve the evolution of a certain complex reaction catalyzed by an enzymaticimmobilized biocatalyst (or a cascade of them), which may be difficult or impossible to performin an aqueous medium. The problem is to find it at a low cost.
