Hepatitis B virus resistance substitutions: long-term analysis by next generation sequencing

Chronic hepatitis B is treated with nucleos(t)ide analogs. Next-generation sequencing (NGS) was applied to analyze longitudinally the resistance-associated mutation (RAMs) patterns from 7 patients at baseline and during therapy. Phylogenetic analysis was applied to infer relationships among the HBV haplotypes. Patients were identified with sub-genotype A2 and F1 infections. Primary antiviral-resistant mutants existed as single variations in untreated patients with very low frequency but during therapy these appeared as multiple ones with higher abundance. In conclusion, using NGS we observed HBV variants exhibiting RAMs before treatment and their dynamics during antiviral treatment is complex could promote NA-resistance under antiviral therapy.
