Production of recombinant enzymes of wide use for research

For biotechnological purposes, protein expression refersto the directed synthesis of large amounts of desiredproteins. The aim of the present work was to producereverse transcriptase Moloney murine Leukaemia Virusretro-transcriptase and Taq DNA polymerase, asbioactive products. In the present paper, we report thepreparation of recombinant enzymes, expressed in E.coli strains. The enzymes produced exhibited quite goodactivity, compared with commercial enzymes, allowingus to replace the last ones for several lab applications.We are reporting changes and modifications tostandard protocols described. The standard protocolswere modified, i.e. for the purification step of Taq, atemperature dependent procedure was designed. Theenzymes produced were used in different applications,such as PCR, RT-PCR, PCR Multiplex and RAPDsmolecular markers.
