Alzheimer?s Disease(AD) is the first cause of dementia, and it represents a serious medicalproblem that severely affects social, sanitary, and economic aspects of population.Late-onset AD (age at onset > 65 years) is the most frequent form. This is acomplex disorder, most likely caused by multiple genetic and environmentalsusceptibility factors. The pathophysiological process of AD is thought tobegin many years before the appearance of dementia symptoms. This preclinicalphase of AD would provide a critical opportunity for therapeutic intervention. Inthis chapter we explore the relationship between AD neuropathology, neuronal bioenergeticsand behavior in transgenic animal models of AD. We compare bioenergeticsparameters obtained from isolated mitochondria and synaptosomes addressingadvantages and disadvantages of each biological preparation. Furthermore, wespeculate to target synaptosomal bioenergetic defect to slow or prevent theneurodegenerative process and restore neuronal function.
