Prenatal exposure to amphetamine (PEA) inducesendurable changes that become manifest in the adulthood. These changes can beexpressed as an altered central dopaminergic activity. Sex steroids modulatebrain dopaminergic systems and may influence the neuroadaptive changes inducedby amphetamine. Stress exposure induces dopamine release in the mesolimbicpathway and the activation of hypothalamic-pituitaryadrenal axis. Our aim wasto evaluate the effect of estradiol in PEA animals in response to stress anddopaminergic receptors and tyrosine hydroxylase (TH) expression in nucleusaccumbens. To this purpose, female rats were treated daily with amphetamine2,5mg/ kg i.p or saline during days 15 to 21 of pregnancy. Their Offspring wereovariectomized (OVX) at day 60, treated with estrogen (E2) (2x 5ug/rat/24 h) oroil 15 days later, and subjected to immobilization stress during 2 h. Blood andtissue samples were collected to determine corticosterone by RIA and dopaminereceptors (D1R, D2R, D3R) and TH expression by real time PCR in extracts ofnucleus accumbens. Stress exposure increased corticosterone levels in OVX (basal:109,19 ±44,57 ng/ml vs stress: 214,41±21,49 ng/ml) and OVX+E2 (basal:157,24±33,87 ng/ml vs stress: 364,04±23,75 ng/ml), in both groups PEA bluntedthe corticosterone response to stress. PEA induced a decrease in basal D3R expression andan increase in TH expression in response to stress, both responses were E2dependent. No change was observed in D1R and D2R expression. We conclude thatthe neuroadaptive response induced by PEA in nucleus accumbens is underestrogen influence. Moreover, an altered stress response should be consideredbetween the neuroadaptive changes induced by PEA.
