GABAB RECEPTOR DELETION IN KISS1 NEURONS/CELLS AFFECTS THE HYPOTHALAMIC EXPRESSION OF KISS1 IN MALE MICE

Resumen:
It is known that Kiss1 cells/neurons coexpress GABAB receptors (GABABRs), and our group haspreviously demonstrated that global GABAB1KO mice show adramatic increase in Kiss1 expression in male and femaleextrahypothalamic areas (amygdala, BNST, lateral septum), butits expression does not differ in AVPV-PeN/ARC. To establish theimpact of GABABRs on kisspeptin physiology we developed astrain of mice with specific deletion of GABABRs in Kiss1cells/neurons and characterized them from reproductiveperspectives. Kiss-Cre mice (Jackson?s Lab) were crossed withGABAB-floxed mice (donated by Dr. Bettler) to obtain Kiss1-GABAB1KO (KO) mice. Male Kiss1 expression was evaluated byqPCR in amygdala, BNST, AVPV-PeN and ARC micropunches;serum hormones were determined by RIA. Conversely toGABAB1KO mice, no significant changes were observed inextrahypothalamic areas [amygdala (fold change): KO= 0.95 ±0.08 (n= 10) vs. WT=0.98 ± 0.08 (n= 9), p ns; BNST (foldchange): KO= 3.36 ± 1.48 (n= 9) vs. WT= 1.05 ± 0.15 (n= 6),p= 0.07].
