Antimicrobial peptides (AMPs) are small molecules that display antimicrobial activity against a wide range of pathogens. In a previous work by using model membranes we studied two related novel peptides, p6 (GLLWKWGWKWKEFLRIVGY) that no show antimicrobial activity and p6.2 (GLL-RKWGKKWKEFLRRVWK) witch exhibited good antibacterial [1]. Besides biophysical techniques on model membranes can provide very detailed information on the interaction of AMPs with membranes, until recently, it was unknown the information predicted was relevant for AMPs activity in real bacteria. In this context, we aimed to unravel the way of action of theses peptide by studying the effect that each peptides was able to induce in Escherichia coli  ATCC 25922 (Gram -) and Staphylococcus aureus ATCC 25923 (Gram +) by using Zeta Potential, absorbance and fluorescence approaches.
