POSSIBLE PROTECTION OF PSEN2 IN SPERMATOGONIAL STEM CELL INDUCED BY WARM TEMPERATURE

Elevated temperature causes degeneration and disappearance of the germ cells in the males of scrotal mammals and the testis of fish, even producing sterile males. However it is known that in a stress response, spermatogonial stem cells (SSC) are protected to apoptosis. In this context the protection of SSC to stress is crucial to the fertility. Psen2 is part of the gamma secretase complex that influences a number of processes as cell to cell transducer signals and has been characterized as anti-apoptotic gene. The purpose of this study was to clarify the participation of psen2 in the heat-induced SSC protection using medaka fish (Oryzias latipes) as experimental model. Two groups of 10 males were reared at 33°C (warm temperature group, HT) and 26°C (control group, NT). At 0, 3 and 10 days, 5 males per group of HT and NT were sacrificed to remove the testicles. Then, testicles were dissected to whole-mount in situ hybridization and RT-qPCR to characterize the psen2 transcript distribution and abundance. After 3 days psen2 is up-regulated in somatic cells at HT, possibly protecting the entire testicle to apoptosis. After 10 days, psen2 transcripts are restricted to somatic cells, surrounding the SSC (identified by oct-4), that could protected the SSCs to the heat-induced degeneration.
