The overexpression of Acyl-CoA synthetase 4 (ACSL4) is involved in the aggressiveness of several cancers, including prostate and breast cancer. It has already been studied that the androgen receptor (AR) expression or androgenic stimulation decreases expression and mRNA levels of ACSL4 in prostate cancer cells. However, the effects of androgenic regulation in breast cancer cells have not yet been described. The aim of this work is to study whether AR and androgens regulate ACSL4 transcription in breast cancer cells. First, we verified the AR mRNA content in our study models. We found high levels of AR mRNA in MCF-7 cell line while in MDA-MB-231 the AR mRNA content is practically zero. In a bioinformatic analysis performed with the Genomatix tool and contrasted with the Alggen Promo software, we found at least one AR element (ARE) in a 1.8 kb fragment of the human ACSL4 promoter with a score close to 1.
