Abstract. The effects of two different voltage-dependent calcium channel (VDCC) blockers, ω-agatoxin IVA (100 nM) and ω-conotoxin GVIA (1-5 µM) were tested on evoked transmitter release at NMJ of diaphragm and androgen-sensitive levator ani muscle of Sprague Dawley male rats. Evoked endplate potentials were recorded intracellularly and quantal content was estimated by the failure method. ω-agatoxin IVA, a P/Q type VDCC blocker, caused a significant reduction in quantal content at levator ani but not at diaphragm NMJ of mature rats. Therefore, this results indicate that neurotransmitter release is mediated by P/Q and N-type calcium channels in the levator ani but not in diaphragm NMJ of the rats. The presence of N-type calcium channels in the levator ani NMJ may be related to the sensitivity of this muscle to sexual hormones.
