Cattle undergo numerous environmental and management stressors, which reducefertility and affect ovulation. The extracellular matrix of the ovarian follicular wallcan be altered by matrix metalloproteinases (MMPs), whose activities areregulated by interleukins and tissue inhibitors (TIMPs), especially during ovulation.The aims of the present study were to evaluate changes in the hormonal milieu aswell as the localization and activity of MMP2 and MMP9 and the localization ofMMP14, TIMP1 and TIMP2 in response to ACTH in the preovulatory period incows, and to determine the direct effect of ACTH on the mRNA expression ofMMP2 and MMP9 in the cultured follicular wall of bovine ovaries obtained from theabattoir. ACTH was administered during proestrus every 12 hours untilovariectomy, which was performed before ovulation. Cortisol levels of ACTH-treated cows were high in plasma and follicular fluid (FF) of preovulatory folliclesrelative to control cows, whereas progesterone presented sub-luteal concentrationsin plasma (p
