The pH sensitive archaeosomes ApH are nanovesicles madeof polar archaeolipids extracted from hyperhalophile archaebacteria,dioleylphosphatidylethanolamine and cholesterylhemisuccinate.+n a previous YorM, Ye have shoYn that ApH efficiently deliver itsaqueous content to the cytoplasm of alveolar macrophages andlung epithelial cells in vitro. /oreover, ApH Yere structurally moreresistant to nebulization than conventional pH sensitive liposomes1.Those features maMe ApH excellent candidates for delivering drugs to the lungs. HoYever, in an in vivo context, inhaled ApH mustfirst interact Yith the pulmonary surfactant 2S lining layer thatcovers the internal surface of the alveolus and provides the loYsurface tension at the airliquid interface. +nteractions Yith the 2Sfilm determine the subsequent retention and translocation of theinhaled ApH and hence their potential activity on target cells. Theinteraction of pH sensitive liposomes Yith 2rosurf, a natural 2S,Yas studiedby measuring changes of pressure and compressibilityat different initial surface pressures after addition of nanoliposomesby injection, in the aqueous subphase, or nebulization onto the2S monolayer. The results obtained shoY that ApH produce thehighest increase of surface pressure in 2S monolayers. This behavioris dependent of the membrane pacMaging and nanoliposomeconcentration, the higher interaction Yas determined Yith a loosy2S monolayer and Yith the highest ApH concentration. Also, theApH incorporation rate into 2S monolayer has been affected bythe Yay of its addition, Yhen it Yas nebulized onto the monolayerthe pressure increase rate Yas higher than Yhen it Yas injected inthe monolayer subphase. Since inhaled ApH Yould interact Yiththe monolayer from the air phase, this second model Yould bemore suitable.
