Sexidentificationis a commonobjectivein molecularecology.Whilemanyvertebratesdis-playsexualdimorphism,determiningthesexcanbechallengingin certainsituations,suchasspecieslackingclearsex-relatedphenotypiccharacteristicsor in studiesusingnon-inva-sivemethods.In thesecases,DNAanalysesserveasvaluabletoolsnotonlyforsexdeter-minationbutalsoforvalidatingsexassignmentbasedonphenotypictraits.In thisstudy,wedevelopeda bioinformaticframeworkforsexassignmentusinggenomicdataobtainedthroughGBS,andhavinganavailablecloselyrelatedgenomeassembledat thechromo-somelevel.Ourmethodconsistsof twoadhocindexesthatrelyonthedifferentpropertiesof themammalianheteromorphicsexchromosomes.Forthispurpose,wemappedRAD-seqlocito a referencegenomeandthenobtainedmissingnessandcoveragedepthvaluesfortheautosomesandX andY chromosomesof eachindividual.Ourmethodologysuccess-fullydeterminedthesexof 165fursealsthathadbeenphenotypicallysexedin a previousstudyand40sealionssampledin a non-invasiveway.Additionally,weevaluatedtheaccu-racyof eachindexin sequenceswithvaryingaveragecoveragedepths,withIndexY prov-inggreaterreliabilityandrobustnessin assigningsexto individualswithlow-depthcoverage.Webelievethattheapproachpresentedherecanbeextendedto anyanimaltaxawithknownheteromorphicXY/ZWsexchromosomesystemsandthatit cantoleratevariousqualitiesof GBSsequencingdata.
