Diseño de un método molecular para la detección de Ilex dumosa en yerba mate elaborada utilizando una secuencia específica ubicada en la región ITS2 del DNA ribosómico

Resumen:
In Misiones two species of genus Ilex prevail: I. paraguariensis and I. dumosa, but only the first one should be usedto manufacture yerba mate products. I. dumosa is frequently used in mixtures with I. paraguariensis, althoughamounts above 1% are considered adulterants. From the technological perspective, it is feasible to differentiatethem through the use of molecular markers such as ITS regions of ribosomal DNA. The aim of this work was tooptimize a molecular technique to detect I. dumosa in yerba mate packages when it could be found as adulterant.Fresh leaves and thirteen trademarks of yerba mate and yerba canchada were used for the experience. For primersdesign ITS regions of I. dumosa were used and amplification conditions were standardized and products weresequenced. The technique sensitivity and specificity were 93.3% and 100% respectively, with a detection limit of1% w / w. The technique was validated through the analysis of commercial and artificially adulterated samples andno traces of I. dumosa were detected in all commercial packages tested.
