Resumen:
It is well known the estrogen effects on pituitary cell growth. Previously we demonstrated the activation of Nrf2 antioxidant pathway in response to DNA damage triggered by 17β-estradiol (E2). Here we analyzed the impact of E2 on p53 and p21 activation and the damage response in pituitary cells. The experimental pituitary tumour was induced by subcutaneous implantation of silastic capsules with estradiol benzoate (30mg) for 10 days (E10) in adult male Wistar rats. Control group was implanted with empty capsules. Then, normal and tumoral pituitaries were collected, with cells being cultured and exposed to E2 (1-10-100nM) for 15, 30 and 60min. The p53 and p21 levels were determined by western blot. To analyze the cell population involved in the oxidative damage response activation, PRL or GH/p-Nrf2 expression was evaluated by immunofluorescence. Statistical analysis: ANOVA-Fischer (p
