Resumen:
Species belonging to C. parapsilosis complex (C. parapsilosis sensu stricto, C. metapsilosis and C. orthopsilosis) are commonly found in different types of yeast micosis as well as in artificial and natural environments. Molecular biology techniques are necessary in order to archive species identification. A common molecular marker for this purpose includes nucleotide sequence named ITS1-5.8s-ITS2 region (ribosomal DNA). The aim of this work is to compare 39 ITS1-5.8s-ITS2 nucleotide sequences of C. parapsilosis complex strains obtained from clinical isolates of Argentinian patients with other sequences that include ITS1-5.8s-ITS2 region of C. parapsilosis complex strains isolated from clinical or environmental sources. 807 nucleotide sequences were retrieved from NCBI`s GenBank (public database) and posteriorly edited and aligned for phylogenetic analysis using Neighbor Joining statistical method in the free software MEGA v7.0.26. C. parapsilosis sensu stricto showed 3 haplotypes, and the other two species of the complex showed 2 haplotypes each. Differences between these haplotypes were SNPs on ITS1 region for C. orthopsilosis, on ITS2 region for C. metapsilosis and on ITS1 region or 5.8s region for C. parapsilosis sensu stricto. This is the first comparative large-scale study of ITS1-5.8s-ITS2 nucleotide region for worldwide samples of C. parapsilosis complex strains against Argentinian clinical strains.
