Background: Reactive arthritis (ReA) may develop after gastrointestinal or genitourinary infections. Dendritic cells (DCs) are central for T lymphocyte activation. We reported chronic Yersinia enterocolitica (Ye)-induced ReA in TNFRp55-/- mice with the participation of IL-12/IL-23 in the inflammatory process. The role of DCs in ReA is unknown. The purpose was to study DCs in mucosa and joint of Ye-infected TNFRp55-/- mice and IL-12/23p40 production in these cells.Methods: C57BL/6 wild-type (WT) and TNFRp55-/- mice were orally infected with 1-5 × 108 Ye O:3. Seven, 14 and 21 days after infection, a single-cell suspension was prepared from mesenteric lymph nodes (MNLs) and regional lymph nodes (RLNs) Total DCs (CD11c+ MHC+) and DC subpopulations were analyzed by flow cytometry. Naïve TNFRp55-/- cells from MLN were stimulated in vitro with PMA/ionomicyn, and the cellular source of IL-12/23p40 was evaluated by intracellular flow cytometry.Result: At day 14 after infection (arthritis onset), TNFRp55-/- mice showed a significant increase in absolute number of the migratory (CD11c+MHChi) and resident (CD11c+MHCint) DCs in both MLN and RLN (p
