Galectin-1 and Yersinia outer protein P regulate innate immune response

The effects of Galectin-1 (Gal-1) in bacterial infection are poorly understood. Here we aimed to investigate the role of Gal-1 and the virulence factor Yersinia outer protein P (YopP) in innate immune responses. Peritoneal macrophages(MØ) from C57BL/6 (WT) and Lgals1-/-(KO) mice were infected with Ye WA-314 or with the yopP deletion mutant (ΔyopP). Uninfected cells were used as control. Moreover, Lgals1-/- MØ were exposed to Gal-1. Nitric oxide (NO), urea, TNF and IL-10 were measured in the supernatants. Gal-1 expression was evaluated by WB. The binding of Gal-1 to Ye WA-314 or ΔyopP was analyzed using flow cytometry and WB. To evaluate the role of NO on Ye clearance in Lgals1-/- mice, they were treated with aminoguanidine (AG) before infection with Ye WA-314. At day 5 after infection, colony forming units (CFU), urea and IL-17 and were measured in Peyer's patches (PP). We observed that in contrast to WT MØ, uninfected Lgals1-/- MØ produced higher amounts of NO and these concentrations were sustained after Ye WA-314 infection (p
