Local expression of cytokine, TLR, Bcl-2 and bax mRNA in yersinia infected IL-12p40-/- mice

Resumen:
Previously we observed that IL-12 deficiency contributes to persistence of Yersinia enterocotitica O:3 in mucosa and  development of reactive arthritis (ReA) on day 21 post-infection (p.i.). The purpose was to investigate in normal C57BL/6 and IL-12p40 deficient (IL-12p40-/-) mice the  local expression of cytokine, TLR, Bcl-2 and Bax mRNA in mesenteric lymph nodes (MLN) and joints. The RNA was isolated from orally infected mice sacrificed on hay 3 or 21 p.i. PCR was performed on cDNA for b-actin, Bcl-2, Bax, TNF-a, IFN-g, IL-10, TLR2 and TLR4. Furthermore, IgG, IgG2a and IgG1 anti-lipopolysaccharide (LPS) O:3, anti-collagen type I and anti-collagen type II in serum were measured by ELISA. In IL-12p40-/- mice, increased expression of TNF-a,  IL-10, TLR-2, TLR4 and Bax mRNA was found in MLN at day 3 p.i. At day 21, TNF-a and Bax mRNA were slightly increased in MNL and joints. IgG anti-collagen type I and type II, and the radio IgG2a/IgG1 anti-LPS O:3 were significantly increased in IL-12p40-/- mice at day 21 (p<0.05). We concluded that ReA in IL-12p40-/- mice may be related with local expression of cytokine and antibody response to LPS and articular collagen.
