Association between high levels IL-8 and Staphylococcus aureus-specific IgA antibodies in subjects with type 1 diabetes mellitus from Argentina.

Interleukin (IL)-8 plays an important role in thechronic vascular complications of diabetes [1,2].Carriage of Staphylococcus aureus is significantlyhigher in diabetics [3]. We studied the associationbetween serum S. aureus-specific IgA and IL-8 veryhigh levels in patients with type I diabetes.Sera from 31 type I diabetic patients and 10 healthysubjects were collected. None of them presentedapparent infectious signs. Autoantibodies GADAband IAA were measured by radioimmunoassay(GAD-AB CIS and IA-AIA Bio International ORISGroup, France) and ICA by immunohistochemistrymethod [4], and IL-8, IFN-g, IL-4, IL-6 and TNF-a byELISA (Pelikine CompactTM, The Netherlands). S.aureus (ATCC 35556) was sonicated (12 pulsed cyclesof 10 s at 20 Hz, Sonics Vibracell) and after centrifugationthe supernatant was used as antigen (SO). Bacterialsecreted proteins (SP) were obtained by overnight47.6% ammonium sulphate precipitation at 4 8C.
