Abstract Ca2+ pump dimerization was studied by using acombined approach of thermal denaturation and fluorescenceresonance energy transfer. The measurement of calcium pumpability to dimerize after the unfolding of individual functionaldomains of the enzyme demonstrated the existence of twodifferent regions involved in the self-association process. One ofthese regions is highly susceptible to thermal unfolding and wasidentified as the calmodulin (CaM)-binding domain. The otherregion whose thermal stability is higher than those of thecatalytic and CaM-binding domains could be related with thepreviously found C28W-binding regions. ß 2000 Federation ofEuropean Biochemical Societies.
