Monodisperse polymer colloids were used as support for their sensitization (by physical adsorption and covalent coupling) with antigenic proteins of Leptospira interrogans. The resulting latex-protein complexes were used in immunoagglutination assays for detecting Leptospirosis by turbidimetry. The approach of the antigenic proteins on the particles surface was favoured at pH close to their isoelectric point. In the covalent coupling experiments, high fractions of proteins were chemically-bound to carboxyl groups on the particles surface and higher densities of linked proteins were obtained with particles exhibiting greater carboxyl groups densities. In the immunoagglutination assays, the latex-protein complexes obtained by covalent coupling with the latex of higher density of carboxyl groups allowed acceptable discrimination between positive and negative bovine sera.
