Resumen:
Excessiveethanoldrinkinghasdeleteriouseffectsonthebrain.However,theeffectsofalcoholonmi-croglia, themainmediatorofthebrain'sinnateimmuneresponseremainpoorlyunderstood.Ontheother hand,theendocannabinoidsystemplaysafundamentalroleinregulatingmicroglialreactivityandfunction. HerewestudiedtheeffectsofacuteethanolexposuretomurineBV2microglialcellson N-acylphosphatidylethanolamine-phospholipase D(NAPE-PLD),amajorsynthesizingenzymeofanandamideand other N-acylethanolamines. WefoundthatethanoldownregulatedmicroglialNAPE-PLDexpressionby activatingcAMP/PKAandERK1/2.Thesesignalingpathwaysconvergedonincreasedphosphorylationof CREB.Moreover,ethanolinducedandincreaseinhistoneacetyltransferaseactivitywhichledtohigherlevels ofacetylationofhistoneH3.Takentogether,ourresultssuggestthatethanolactionsonmicroglialNAPE-PLDexpressionmightinvolveepigeneticmechanisms.
