Resumen:
The aim of this study is to develop a chemical-enzymatic process to produce the high-value chemical 4-hydroxy-2,5-dimethyl-3(2H)-furanone (HDMF). HDMF is present in many fruits, and due to its attractive sensory properties, it is highly appreciated by flavours and fragrances industry. The synthesis of HDMF starts with the aldol condensation of hydroxyacetone and methylglyoxal to form 3,4-dihydroxyhexane-2,5-dione. This product can be obtained by both chemical and enzymatic catalysis. In recent years, many aldolases have been studied as catalysts in the asymmetric synthesis of complex polyfunctional carbohydrates and amino acids. Therefore, in this work a strategy was developed to recombinantly produce an aldolase presents in Escherichia coli which accepts these substrates in vitro. The enzyme was cloned and partially purified by a thermal process. The aldolase activity of the recombinant enzyme was measured with the physiological substrates. Finally, the enzyme was tested in the aldol condensation reaction with the industrial substrates to obtain the HDMF precursor compound.
