Resumen:
A series of intraspecific crosses has been undertaken between different accessions of Olea europaea subspecies europaea and cuspidata with the aim to incorporate traits related to biotic resistance in the hybrids. O. europaea subsp. europaea cultivars Leccino (LE), Frantoio (FR), Dolce Agogia (DA) and FS17, and three Asiatic and African accessions of subsp. cuspidata were crossed to obtain genotypes showing potentially useful trait combinations. Several parent combinations, each involving one cultivar and one cuspidata accession, were exploited. Flowers of seed-bearing parent were emasculated and dusted with pollen of the male parent. Pollination was performed on potted plants in a controlled environment with high relative humidity (60% RH). Viable fruits were obtained from the following parent combinations: a) cuspidata South Africa x FR, b) cuspidata South Africa x LE, c) cuspidata China x DA, d) DA x cuspidata South Africa, e) DA x cuspidata China, f) DA x cuspidata India and g) FS17 x cuspidata India. Embryos were dissected from mature fruits, sterilised with 10% sodium hypochlorite, rinsed with abundant sterile water and allowed to germinate in vitro. The embryo culture was performed in test tubes containing agar-solidified EZ medium and incubated in a growth chamber with fluorescent light. After 60 days of culture, the explants were in vitro micropropagated and later on they were moved to pots in the greenhouse. After appropriate acclimatation well grown plants were obtained. SSR markers were used to investigate the transmission of paternal alleles into the genome of the putative hybrids. Three microsatellites (DCA-09, DCA-05, GAPU103A) were able to detect the presence of the male parent genome in the seedling?s genome thus confirming their hybrid nature.
