Resumen:
Life expectancy has increased exponentially over the last 50 years.This extended lifespan has led to the emergenceofage-relateddisorders,withdementiatakingcenterstage.Amongthevariousformsof dementia,Alzheimer´sdisease(AD)standsoutasthemostcommon,accountingfor60-70%ofcases. Currently, there isnodefinitivetreatment for thiscondition,andalldrugsandinterventions focusing solelyon symptomalleviation, primarily throughcholinesterase inhibitors andmemantine. In this context, scientistshaveembarkedonaquest touncoveragentsorcombinationof themcapableof forestallingorenhancingthequalityof lifefor thoseaffectedbythiscondition. Recent researchhas spotlightednutritional interventions andtheburgeoning fieldof probiotics, whichholdpromise for mentalhealth. Inthepresentstudy,weevaluatedtheeffectofdailyoraladministrationoftwoprobiotic strains: Lactobacillusdelbrueckii subsp. lactisCRL581 (1x108),aninvitroacetylcholinesterase(AChE) inhibitor,andLevilactobacillusbrevisCRL2013(1x109),anefficientgamma-aminobutyricacid(GABA) producing strain. This administrationspannedaperiodof 30daysandaimedto investigate their impact onoxidativestressandcholinergicdysfunctionwithinascopolamine-inducedmicemodel. Scopolamine,actingasacholinergicreceptorantagonist, inducedmemoryloss,cognitiveimpairment, andelevatedAChEactivity, thereby replicatingsomepathologicalalterationsobservedinAD.AChE activity exhibiteda significant increase inbrainhomogenates ofmice treatedwith scopolamine compared to thecontrol group. Bothprobioticstrainsdemonstratedtheability todecreaseAChE activity, withCRL581 showingparticularlynoteworthyefficacy. Furthermore,CRL2013administration increasedcatalaseactivity inmicebrains. Infemales,solelytreatmentswithCRL581strainshoweda decreaseinMDAlevels,anend-productof lipidperoxidation,whereasinmales,bothstrainsdisplayed thecapacitytoreduce it.NosignificantdifferenceswereobservedinGSHactivityacrossallgroups. Data fromtheshotgunproteomicof scopolamine-andpsychobiotics-treatedbrainhomogenates revealeddistinctlyanduniquedifferential expressionpatterns ineachgroup. Thesefindingsstrongly support the potential development of a functional supplement using t, offering a promising non-pharmacological interventionforindividualsaffectedbyAlzheimer´sdisease.
