Free radicals are implicated in several health diseases, such as
Parkinson's disease, Alzheimer disease, diabetes, cancer, etc. Our
previous reports showed that the infusion prepared from T.
acutifolius leaves has free radical scavenging activity. The aim of
this study was to perform a biodirected isolation and identification
of the antioxidant substances from T. acutifolius infusion. Lyophilized
Infusion was extracted with solvents of increasing polarity.
Methanolic fraction was separated by Silica Gel column chromatography
and Sephadex LH20. Final purification was achieved
by preparative TLC developed with ethyl acetate/methanol/water
(1 00: 13.5: 10 vlv) as mobile phase. During development of the purification
method all steps were monitored by qualitative TLC, and
different staining procedures (Vainillin/H2SO4, NP/PEG, and DPPH
for antioxidant activity). Rutin, Iso-quercitrin and Hyperoside were
isolated and identified by comparison of their UV spectra and chromatographic
profiles with those from pure standards. These Glycoflavonoids are responsibles of T. acutifolius infusion antioxidant activity.
