ERYTHROCYTE SENESCENCE MARKERS AS PARAMETER OF TOXICITY OF FUMONISIN

Fumonisin B1(FB1) is a mycotoxin that occurs as a frequent contaminant of corn and corn-based foods in Argentina, has an inhibitoryeffect on ceramide synthetase, key enzyme in the biosynthesis ofsphingolipids. The effects of FB1 on the plasma membrane and onoxidative stress led us to propose a possible role of FB1 on the aging of erythrocytes (EC). Our objective was to assay techniquesto determine the aging of EC to apply to the exposure study by fumonisins. Materials and Methods: Male 1-month Wistar rats wereseparated into two groups of 1 animal. The control rat was givensaline solution while the other rat received orally for 20 days 1 mg/kg/day of FB1 from a culture of Fusarium proliferatum. Erythrocytemalondialdehyde (MDA) was measured according to the method ofKumar et al. The binding of alcian blue (AB) to human or rat EC, tomeasure the effect of fumonisin on erythrocyte charge, was studiedby the method of Ponce de Leon et al. In vitro experiments, EC 5%in PBS was added to three tubes (0, 50 and 100 ng of FB1), incubated at 37 º 24 hs.
